Lonza's bone marrow stromal cells should be thawed according to Lonza's "Procedure for thawing Poietics™ Cells".
Upon thaw, cells should be plated at 3,000-4,000 cells per cm² in MyeloCult™ H5100 from StemCell Technologies, Inc. An adherent monolayer will be established in 3-4 days. At that time, hematopoietic progenitors can be added to begin an LTC-IC assay. The stromal layer should be capable of supporting hematopoiesis for up to 8 weeks.
