Q:

pmaxGFP™ – the control plasmid in our Nucleofector® Kits

A:

The pmaxGFP™ plasmid included in all our Nucleofector® Kits is intended for use as a control only. This control vector is under license from Evrogen.

We are not allowed to share the complete sequence. However, we can provide you a simple vector map which might be helpful.

Please note:

-        The plasmid size is about 3,5 kb.

-        The coding sequence for the GFP ends with a stop codon (TGA/UGA).

-        The molecular weight of the fluorescent protein is about 27kDa.

-        The codon optimized backbone is a variant of the MIDGE vector system.

-        This vector cannot be used for selection because it does not contain a mammalian selection marker. The only resistance gene expressed by this vector is kanamycin which is only suitable for bacterial selection.

-        Unlike common GFPs from Aequorea victoria, pmaxGFP™ originates from Pontellina plumata and requires different antibodies for detection:

Evrogen AB513 for the denatured form ( Evrogen Anti-TurboGFP(d) antibody (CAT.# AB513) )

-        The pmaxGFP™ vector provided in our Nucleofector® Kits is not expressed in insect cells.
