Q:

Why does Lonza recommend two different concentrations of the B-Glycerophosphate in the Normal Human Osteoblasts (NHOst) differentiation protocol?

A:

Lonza R&D has found some NHOst cell strains to be more sensitive to B-Glycerophosphate than others.

While it is necessary to differentiate, at too high of concentrations the cells will lift off dish, curl up and die.

As such, Lonza recommends trying these two cited concentration to determine the optimal concentration for each specific lot.
