Lonza currently validates trilineage differentiation of human mesenchymal stem cells using the following quantitative assays:

  • Adipogenic Differentiation: lipid vacuole binding with AdipoRed (catalog no. PT-7009)
  • Osteogenic Differentiation: inorganic hydroxyapatite binding with OsteoImage (catalog no. PA-1503)
  • Chondrogenic Differentiation: staining with alcian blue

Please note that other methods including Oil Red O staining for adipogenic differentiation, ELISA assays for calcium for osteogenic differentiation, and Safranin staining for chondrogenic differentiation have been used in the past and are all acceptable methods for validating differentiation.
