What is the difference between each of the types of HUVECs that Lonza offers?

When choosing between the various types of HUVECs that Lonza offers, the choice can be made by answering the following questions:

  • What media do you intend to culture the cells in?
  • Do you require pooled or single donor cells?
  • Do you require additional screening for angiogenesis and VEGF markers?
  • Do you intend to use the cells in a high-throughput assay?

What media do you intend to culture the cells in?  Lonza offers HUVECs isolated in both the original EGM™ Media as well as the newer EGM™-2 Media. Cells tend to proliferate better in the EGM™-2 Media, however some customers have indicated that the original EGM™ Media works better for VEGF, FGF and angiogenesis research. Cells which have been cultured in the original EGM™ Media can be immediately transitioned into the EGM™-2 Media at any time, but once cells are cultures in the EGM™-2 Media, they cannot be transitioned back to the EGM™ Media.

Do you require pooled or single donor cells? Lonza offers HUVECs from both pooled and single donor sources. Single donor HUVECs are typically suggested for studies where a specific donor characteristic (race, gender, etc) is necessary. Pooled HUVECs are generally suggested for most other applications as the nature of the pooled product limits the potential for lot-to-lot variability.

Do you require additional screening for angiogenesis and VEGF markers? Lonza offers HUVECs which have received additional product testing and are guaranteed to test positive for angiogenesis and vascular cell health markers, eNOS, Tie-2, VEGFr2, and Axl in the first passage out of cryopreservation. These cells have undergone the same testing as the non-screened cells and will be guaranteed for the same testing specifications as the non-screened cells (viability, population doublings, plating efficiency, markers, etc) in addition to the additional screening tests.

Do you intend to use the cells in a high-throughput assay? Lonza offers HUVECs which have been pre-expanded and cryopreserved in high volumes making them appropriate for high throughput assays. These HUVECs are created using our pooled, non-screened HUVECs. The cells are expanded to Passage 3 and cryopreserved at 10 million cells per vial (all other HUVECs are expanded to Passage 1 and cryopreserved at 500,000 cells per vial). The intention for this product is for the researcher to thaw the cells and plate them into their high-throughput assay format immediately from cryopreservation eliminating the time needed to otherwise expand an ampoule of cells prior to assaying. These cells are only guaranteed for 5 population doublings (all other HUVECs are guaranteed for 15 population doublings) and therefore it is not suggested to attempt to further expand, subculture, or bank these cells after thawing.
