Do you recommend a purification method to purify specific blood/immune cell populations before Nucleofection®?

A:

We recommend using negative selection (depletion) because your purified cells are "untouched", not influenced, and strictly not activated.

Positive selection may cause increasing amounts of dead cells and could also lead to activation of the cells due to binding to a specific ligand. 

We made good experience with the MACS™ Selection Kit (Miltenyi Biotec GmbH), that results in a high purification (up to 97-98%) of the specific cell population.

Alternatives are RosetteSep™ Kits (StemCell Technologies Inc.) or Invitrogen™ Dynabeads™(Fisher scientific).
