The pmax-GFP control vector is under license from Evrogen. We are not allowed to share the complete sequence. We can provide you a simple vector map which might be helpful. If you need only the GFP sequence, please contact Scientific Support.
The control vector does not have a multicloning site and no mammalian resistance gene, the codon optimized backbone is a variant of the MIDGE vector system.
Our control vector is made by Lonza with a codon optimized backbone from the Midge vector.
Minimal size MIDGE vectors improve transgene expression in vivo
F Schakowski, M Gorschlueter, P Buttgereit, A Maerten… - in vivo, 2007 - iv.iiarjournals.org
Viral and plasmid vectors may cause immunological side-effects resulting from the expression of therapeutically unwanted genes and from CpG motifs contained in their sequence. A new vector type for minimalistic, immunological-defined gene expression …
In this backbone we cloned with Evrogen licence the copGFP sequence. The copGFP is not suitable for long term expression, in some specific cell types like neurons it builds aggregates which might be toxic for some cells. The further development of the copGFP is the turbo GFP.
Lonza has discontinued the pmax-Cloning vector and the FP green and red expression vectors.
For the cloning I would recommend to use the multicloning vector with strong expression and MSC pTURBOGFP-C
